- 0:05it is always important to follow best
- 0:08practices while working in a biosafety
- 0:10cabinet also known as a BSC these
- 0:14practices will help maintain adequate
- 0:16airflow within the cabinet to help
- 0:19prevent exposures and release of
- 0:20pathogens they should also be
- 0:22incorporated into your facility's
- 0:24standard operating procedures now let's
- 0:28take a look at some of these recommended
- 0:29practices work in a BSC should be
- 0:32carried out by one person at a time all
- 0:35work in a BSC should be carried out as
- 0:38far as possible towards the rear of the
- 0:40work area elbows and arms should not
- 0:43rest on the grill or work surface unless
- 0:46the BSC has been outfitted with
- 0:48appropriate arm rests one should avoid
- 0:51quick and excessive movement of hands
- 0:53and arms through the front opening such
- 0:55movements disrupt the air curtain at the
- 0:58front of the BSC
- 0:59which can allow contaminants to enter or
- 1:01escape arms should enter and exit the
- 1:04BSC slowly and perpendicular to the
- 1:07front opening a bottle of an appropriate
- 1:10disinfectant should be kept within the
- 1:12BSC while work is being performed this
- 1:16will reduce the need to remove your
- 1:17hands from the BSC it is recommended to
- 1:21segregate non contaminated items also
- 1:24known as clean items from contaminated
- 1:27items also known as dirty items work
- 1:30should always flow from clean to dirty
- 1:32areas material should be discarded in a
- 1:36waste container that is located towards
- 1:38the side of the cabinet workspace do not
- 1:42discard materials in containers outside
- 1:44of the cabinet open flames such as
- 1:47Bunsen burners should not be used inside
- 1:50a BSC as they create turbulence disrupt
- 1:53airflow patterns and can damage the HEPA
- 1:56filter if absolutely necessary touch
- 2:00activated micro burners are preferred to
- 2:02reduce airflow disturbance non flame
- 2:06alternatives such as sterile disposable
- 2:08inoculation loops
- 2:10be used whenever possible equipment
- 2:13creating air movement such as vacuum
- 2:15pumps and centrifuges may affect the
- 2:18integrity of the airflow and should not
- 2:20be used within the BSC and finally
- 2:23windows should be kept closed and
- 2:25cooling fans should be turned off when
- 2:27the BSC is in use in order to minimize
- 2:30disruptions to the air movement once you
- 2:33have completed your work you still have
- 2:35a few additional procedures to follow
- 2:37before turning off the BSC containers
- 2:41should be closed or covered and all
- 2:43items should be surface decontaminated
- 2:46before removing them from the BSC
- 2:48disinfect the interior surfaces of the
- 2:51BSC including sides back and interior of
- 2:55the glass use a disinfectant that is
- 2:58effective against the pathogens in use
- 3:00allowing an appropriate contact time if
- 3:03a corrosive disinfectant is used the
- 3:06surface should be wiped down after
- 3:08disinfection and sprayed with 70%
- 3:10ethanol to avoid corrosion of the
- 3:13stainless steel surfaces one should also
- 3:16routinely remove the work surface and
- 3:18disinfect the tray beneath you should
- 3:21also allow sufficient time for the air
- 3:23in the BSC to purge before disrupting
- 3:26the air curtain by removing hands or
- 3:28unloading material from the BSC the
- 3:31purge time will vary by model and can be
- 3:34up to several minutes the use of
- 3:36ultraviolet UV germicidal lamps to
- 3:39disinfect a contaminated BSC is strongly
- 3:42discouraged due to their limited
- 3:44effectiveness at disinfecting the inside
- 3:47of BSC s UV irradiation of the work area
- 3:51should only be used as a secondary
- 3:53method of disinfection in the cabinet
- 3:56let's conclude this video with a review
- 3:59of the following key messages a facility
- 4:02standard operating procedures should
- 4:04include best practices that minimize the
- 4:06disruption of air flow while working in
- 4:09a BSC
- 4:10work from clean to dirty and dispose of
- 4:13contaminated waste inside the cabinet it
- 4:16is important to decontaminate items and
- 4:19surfaces within the BSC after completing
- 4:22your work do not rely on UV irradiation
- 4:26alone to disinfect a contaminated work
- 4:29area knowledge of how to work properly
- 4:32in a BSC is a key component to working
- 4:35safely when using them
