  3. 0:08containment devices designed to protect
  4. 0:10laboratory workers from potential
  5. 0:12exposure to infectious agents and the
  6. 0:15release of the agents to the surrounding
  7. 0:16environment prior to working with
  8. 0:19infectious material inside the biosafety
  9. 0:21cabinet there are a few important steps
  10. 0:24to take to make sure that work will be
  11. 0:26done safely and the cabinet is
  12. 0:27functioning correctly every institution
  13. 0:30should develop a standard operating
  14. 0:32procedure or SOP for the correct use of
  15. 0:35biosafety cabinets the SOP should
  16. 0:38contain introductory information on BSC
  17. 0:41s outline the risk assessment process
  18. 0:43for selecting the most appropriate
  19. 0:45cabinet outline staff training
  20. 0:48requirements and provide detailed
  21. 0:50information on all steps required for
  22. 0:52the safe use of this equipment before
  23. 0:55using a biosafety cabinet laboratory
  24. 0:58staff should verify the most recent
  25. 1:00certification date recertification is
  26. 1:03ideally done on an annual basis however
  27. 1:05many parts of the world lack trained
  28. 1:08technicians and equipment resulting in
  29. 1:10BSC s that are serviced less frequently
  30. 1:12during periods between recertification
  31. 1:16there are a few simple ways that
  32. 1:17laboratory staff can perform qualitative
  33. 1:20assessments of the error curtain and
  34. 1:21function of the HEPA filter holding a
  35. 1:24lightweight tissue paper and moving over
  36. 1:26the areas immediately in front of the
  37. 1:29opening the paper should always be
  38. 1:31pulled inward this provides a visual
  39. 1:33confirmation that the air curtain is
  40. 1:35being maintained a similar procedure can
  41. 1:38be performed inside the cabinet by
  42. 1:40grasping a tissue paper with a pair of
  43. 1:42tweezers and moving it around the
  44. 1:44cabinet to observe airflow patterns if
  45. 1:47unexplained contamination has become a
  46. 1:50recurring problem the HEPA filter
  47. 1:52responsible for delivering sterile air
  48. 1:54over the work surface could also be a
  49. 1:56contributing factor a damaged or leaky
  50. 1:59supply HEPA can blow unfiltered air over
  51. 2:02the work surface as a qualitative test
  52. 2:05place open agar plates in the biosafety
  53. 2:08cabinet for a period of time and check
  54. 2:10for bacterial growth following
  55. 2:12incubation
  56. 2:13the presence of colonies on the plate
  57. 2:15could indicate a serious problem with
  58. 2:17the BSC however it's important to
  59. 2:20remember that not everything the heba
  60. 2:22serves to filter out will grow on an
  61. 2:24agar plate and lack of growth does not
  62. 2:27guarantee that the filter is functioning
  63. 2:29as required for these reasons
  64. 2:32recertification by trained technicians
  65. 2:34with the appropriate equipment is the
  66. 2:36only way to ensure that the critical
  67. 2:38components of a biosafety cabinet are
  68. 2:40functioning as required the first step
  69. 2:43before using a biosafety cabinet is
  70. 2:45turning on the blower allow sufficient
  71. 2:48time for the air to purge and the
  72. 2:50pressure reading to stabilize before
  73. 2:52initiating work this will be specified
  74. 2:55in the manufacturer's instructions and
  75. 2:57is generally three to five minutes
  76. 2:59ensure that the air intake and exhaust
  77. 3:02grills are free from obstructions and
  78. 3:04confirm inward airflow with a tissue
  79. 3:07prior to each use based on your local
  80. 3:10risk assessment additional personnel
  81. 3:12protective equipment or PPE may be
  82. 3:15required for the work you'll be
  83. 3:16performing tuck the cuffs of your gown
  84. 3:19or lab coat into your gloves to reduce
  85. 3:21the likelihood of having exposed skin
  86. 3:24lab coats with elastic cuffs are
  87. 3:26preferred over those with standard
  88. 3:28sleeves as there is less of a chance
  89. 3:30they will slip out of your gloves and
  90. 3:31potentially come in contact with your
  91. 3:33work next clean the interior surface of
  92. 3:37the cabinet with 70% ethanol or other
  93. 3:39appropriate disinfectant wear gloves to
  94. 3:42protect your skin once the BSC is clean
  95. 3:46work materials may be placed inside to
  96. 3:49facilitate clean up later on plastic
  97. 3:51packed absorbent towel may be placed on
  98. 3:53the work surface disinfect items with
  99. 3:5670% ethanol before placing them inside
  100. 3:59to avoid introducing potential
  101. 4:01contaminants into the cabinet try to
  102. 4:04bring in all the materials you'll need
  103. 4:05without overcrowding the cabinet and
  104. 4:08ensure adequate supplies for elimination
  105. 4:10of waste and disinfection while working
  106. 4:13while 70% ethanol is highly effective
  107. 4:16against many pathogens a
  108. 4:19assessment will help you select the best
  109. 4:20disinfectant to keep on hand as a
  110. 4:23practical example will prepare the
  111. 4:25cabinet for processing patient samples
  112. 4:27for PCR and other downstream testing
  113. 4:30arrange the items inside the BSC in a
  114. 4:33logical order that allows work from
  115. 4:35clean to dirty roughly dividing the
  116. 4:38cabinet workspace into three sections
  117. 4:40keep clean items towards one outer end
  118. 4:43designate the middle third of the
  119. 4:45cabinet as a work zone and keep waste
  120. 4:47receptacles at the other end aerosol
  121. 4:50generating equipment such as vortex
  122. 4:52mixers should be placed as far to the
  123. 4:55rear of the cabinet as possible
  124. 4:57taller items such as biohazard bags
  125. 5:00should be placed towards the side walls
  126. 5:02to minimise airflow disruption for this
  127. 5:05reason trays for contaminated
  128. 5:06serological pipettes are preferred over
  129. 5:09vertical waste receptacles next place
  130. 5:12the biological materials inside the
  131. 5:15cabinet once everything is inside the
  132. 5:17BSC adjust the chair height so that
  133. 5:20armpits are level with the sash opening
  134. 5:22you are now ready to begin your work
  135. 5:25let's conclude this video with a review
  136. 5:28of the following key messages ensure BSC
  137. 5:32s are adequately serviced and maintained
  138. 5:34prior to use don appropriate PPE
  139. 5:38disinfect the inside of the cabinet and
  140. 5:40the surface of all items placed inside
  141. 5:43organise materials to promote workflow
  142. 5:46from clean to dirty keep a supply of
  143. 5:49disinfectant and cleaning materials
  144. 5:51inside the cabinet safe use of a BSC
  145. 5:55begins even before sitting at the
  146. 5:57cabinet for work planning ahead and
